Enzymatic Digestion of Desmosome and Hemidesmosome Plaques Performed on Ultrathin Sections
نویسندگان
چکیده
The fine structure of desmosomes is well established (1-3), and some information about their chemical composition is available (4-6) . Recently techniques were developed for performing enzymatic digestions on osmium tetroxide-fixed, alcoholor acetone-dehydrated and epoxyembedded tissues (7-10) . These methods can determine with considerable specificity the organic components of many cell structures (8-10) . Protease digestion, for example, permits chemical discrimination among certain morphological components of desmosomes and hemidesmosomes .

Isolation of the non-glycosylated proteins of desmosomes and immunolocalization of a third plaque protein: desmoplakin III.
The cytoplasmic plaque of the spot desmosome or macula adhaerens mediates the attachment of bundles of intermediate filaments to the plasma membrane. We have isolated from a bovine epidermal desmosome preparation a fraction that is highly enriched in the non-glycosylated desmosomal proteins. Plastic-embedded and thin-sectioned high-speed pellets of this fraction reveal closely packed filaments ...
متن کاملIntracellular Characterization of Bean Yellow Mosaic Virus-induced Inclusions by Differential Enzyme Digestion
The composition of crystalline inclusions and dense bands in cells of broad bean leaves infected with bean yellow mosaic virus was studied by differential enzymatic digestion. Frozen thick sections were prefixed in glutaraldehyde and exposed to proteinases and nucleases, after which ultrathin sections were prepared for electron microscopy. Examination revealed that the crystals were completely ...
متن کاملIdentification of actin-, alpha-actinin-, and vinculin-containing plaques at the lateral membrane of epithelial cells
In this paper, a new type of spot desmosome-like junction (type II plaque) is described that is scattered along the entire lateral plasma membrane of rat and human intestinal epithelium. Ultrastructurally type II plaques differed from the classical type of epithelial spot desmosome ("macula adherens", further denoted as type I desmosome) by weak electron density of the membrane-associated plaqu...
متن کاملIsolation of Epidermal Desmosomes
A method is reported for the isolation of desmosomes in a high yield and of a purity suitable for biochemical analysis. The procedure utilizes the selective solubilizing action of citric acid-sodium citrate (CASC) buffer, pH 2.6, on the non-cornified layers of cow nose epidermis, followed by discontinuous sucrose density gradient centrifugation. Electron microscopy with both thin sections of pe...
متن کاملDifferent isolation methods of dental pulp stem cells
Considering the ease of isolation and high expansion potential of pulp stem/progenitor cells isolated from wisdom and primary teeth they have been implicated as the most reliable autologous cell source in dental tissue engineering. Meanwhile, different isolation methods have remarkable impacts on the expansion potential of adult stem cells. In enzymatic digestion method extracted teeth as denta...
